Effect of subacute ruminal acidosis on milk fat concentration, yield and fatty acid profile of dairy cows receiving soybean oil

The objective of this study was to investigate the effect of ruminal infusion of soybean oil (SBO) with either a moderate- or high-forage diet on fat concentration, yield and composition in milk from dairy cows. Six rumen-fistulated Holstein dairy cows (639±51 kg body weight, 140±59 days in milk) were used in the study. Cows were randomly assigned to one of two dietary treatments, a high forage:concentrate (HFC, 74:26) or a moderate forage:concentrate (MFC, 56:44) total mixed ration. Cows were fed at 08.00 and 13.00 h and pulse-dosed ruminally at 13.00 h over a 10-min duration with 2% of diet dry matter of SBO. Ruminal pH was recorded continuously. Cows receiving the MFC treatment had lower daily mean ruminal pH and ruminal pH was below 6·0 for a longer duration compared with the HFC treatment (640 vs. 262 min/d, P<0·05). Cows receiving the MFC treatment had a greater reduction (diet by week interaction, P<0·05) in milk fat concentration and yield than cows receiving the HFC treatment (42 vs. 22% and 45 vs. 21%, respectively). Additionally, cows receiving the MFC diet had a greater reduction in milk fat concentration (g/100 g FA) of FA <C16 (14 vs. 8%), and a greater increase in concentration of FA >C16 (17 vs. 9%), trans-10 18:1 (159 vs. 21%) and trans-9, cis-11 conjugated linoleic acid (121 vs. 55%) (P<0·05) compared with cows receiving the HFC diet. This study demonstrated that cows fed the MFC diet had lower ruminal pH and showed a greater rate of milk fat depression when infused with SBO.

The role of the gastrointestinal tract (GIT) in explaining the less efficient utilization of metabolizable energy (ME) in growing lambs fed forage rather than concentrate-based diets was investigated by feeding forage (legume–grass silage) and concentrate (whole shelled maize) diets, at isoenergetic intakes (ME basis), using five groups of lambs. One group of seven lambs was an initial slaughter group and of the two groups (eight lambs per group) fed each diet, one group was fed for 8 weeks, whereas the other group was fed for 16 weeks. All lambs were slaughtered between 18·5 and 20 h following their last meal. Retained energy (as a percentage of ME intake) was higher (concentrate-fed 28, forage-fed 17; P<0·001) for the concentrate-fed animals. Weight-specific mucosal O2 uptake (ml/g DM per h), measured in vitro, was 37 % higher for the forestomach (reticulum, rumen and omasum) and small intestine (jejunum) than for the abomasum and large intestine (caecum and colon), but there was no evidence for a diet effect (except colon; forage-fed 5·3, concentrate-fed 4·2; P=0·036). Total GIT heat loss was estimated as 14 (forage-fed) and 18 (concentrate-fed) % of the whole-body heat loss. Although the GIT did not contribute to increased thermogenesis in the forage-fed lambs in the present study, greater relative contribution of GIT tissue to whole-body mass, i.e. GIT as a percentage of empty-body weight(forage 7·6, concentrate 6·6; P<0·001) in the forage-fed animals supports a role for the GIT in contributing to higher thermogenesis observed in ruminants fed forage as opposed to concentrate diets.

A dynamic mathematical model of blood flow regulation in the mammary glands of a lactating cow was constructed from a principle of local vasodilator release in response to changes in intracellular adenylate charge. An equation was derived to predict uptake of the milk precursors acetate, glucose, amino acids and fatty acids, as affected by mammary blood flow rate. Metabolism of the precursors to milk components and CO2 was simulated with a set of empirically derived equations. Relative rates of ATP production and utilization regulated both the number of perfused capillaries and the conductance of arteriolar segments in the mammary glands. The model simulated local control phenomena of functional and reactive hyperaemia, and simulation of autoregulation under changing arterial pressure suggested a predominance of precapillary sphincter regulation. It was predicted that an increase in blood flow without the mammary capacity to utilize blood metabolites efficiently would be detrimental to milk synthesis. Conversely, increased blood flow through changes in mammary activity resulted in predictions of higher milk production. It was proposed that the equation for uptake,
be used in analysis of mammary arteriovenous differences.
Energy expenditure associated with sodium/potassium transport and protein synthesis in skeletal muscle and isolated hepatocytes from hyperthyroid sheep

The object of the present study was to determine the effect of thyroxine (T4) treatment of sheep on protein synthesis and associated energy costs in skeletal muscle and hepatocytes. Protein synthesis, and ouabain-sensitive and cycloheximide-sensitive respiration in isolated intercostal muscle and hepatocytes were determined in sheep after 5 weeks of daily injections of either saline or T4. Plasma T4 and total triiodothyronine (T3) concentrations were doubled and free T3 concentrations were quadrupled by T4 injections. The fractional rates of protein synthesis increased in isolated external intercostal muscle and hepatocytes from hyperthyroid sheep. Fractional rates of protein synthesis in isolated external intercostal muscle and hepatocytes were linearly correlated with plasma free T3 concentrations. Total oxygen consumption of muscle and hepatocytes was unaffected by T4 injections. Ouabain-sensitive respiration increased in hepatocytes and muscle of T4-treated animals. Cycloheximide-sensitive respiration was elevated in hepatocytes from hyperthyroid sheep. Cycloheximide-sensitive respiration in muscle was unaffected by T4 treatment. The present experiment demonstrates that T4 increases protein synthesis in ruminants. The energy expenditure in support of Na+, K+-ATPase and protein synthesis in skeletal muscle and hepatocytes may account for 34–60% of total cellular energy expenditure.
